GLP Toxicology Study Checklist

Every field optionalPlanning checklist, not a compliance checkNothing you type is uploaded

The GLP Toxicology Study Checklist turns what you know about a program into a planning checklist for an IND-enabling toxicology package: modality, indication, stage, route, proposed species, duration, target milestone, test article availability, known requirements and your questions for the CRO. It does not decide which studies are required or establish compliance; it makes sure the right questions are asked.

Every field is optional. Leave a field blank and the plan says Not provided; tick Not decided yet and it says To be decided. Both go on the checklist of open decisions.

Also confirm

  • Which studies are needed, and in what order, decided with a regulatory expert (this checklist does not decide that)
  • Dose-range-finding data available to set GLP dose levels
  • A bioanalytical method for toxicokinetics validated before the GLP study starts
  • Test article characterization and stability documentation ready for the study file
  • Whether a pre-IND or scientific advice meeting is planned before the design is fixed
  • The CRO's GLP compliance status and most recent inspection outcome

This outline organizes requirements you entered. It is not a validated protocol, a sample size recommendation or a regulatory assessment. Have it reviewed by qualified scientific, statistical, ethical and regulatory experts before any study starts.

What is the difference between GLP and non-GLP toxicology studies?

Good Laboratory Practice is a quality system for how nonclinical safety studies are planned, performed, monitored, recorded, archived and reported. In the US, nonclinical studies that support applications to FDA follow 21 CFR Part 58; elsewhere, studies follow the OECD Principles of Good Laboratory Practice. Pivotal toxicology studies that support first-in-human trials are normally run under GLP. Earlier studies, such as dose range finding, are usually non-GLP and are used to choose the doses for the GLP studies.

Which guidelines shape the toxicology package?

Which studies a program needs is a regulatory decision for each program, made with a regulatory expert. These ICH guidelines frame that decision:

  • ICH M3(R2): clinical trials are generally supported by repeated-dose toxicity studies in two mammalian species, one a non-rodent, at least as long as the planned trial
  • ICH S6(R1): for biologics, studies use pharmacologically relevant species; two are normal, but one can be enough in justified cases
  • ICH S9: a streamlined nonclinical package for drugs intended for patients with advanced cancer
  • ICH S7A: the safety pharmacology core battery covers the cardiovascular, respiratory and central nervous systems
  • ICH S2(R1): genotoxicity testing

What should be ready before you contact a toxicology CRO?

Test article is the usual bottleneck: how much is available, from which batch, how well it is characterized, and how long it is stable. A validated bioanalytical method is needed for toxicokinetics before the GLP study starts. Dose range finding data should exist to set the GLP doses. Know the clinical route, regimen and planned trial duration, because the toxicology design follows them, and decide whether a pre-IND or scientific advice meeting comes before the design is fixed.

For timing and budget, the IND Timeline Planner and the GLP toxicology cost guide use the same program inputs. For a plain overview of the whole package, read what IND-enabling studies are at guides what are ind enabling studies.

Frequently asked questions

Does this checklist tell me which toxicology studies are required?

No. Which studies a program needs, and in what order, is decided with a regulatory expert for that program. The checklist organizes what you know and the questions to ask; it does not establish GLP or regulatory compliance.

What is the difference between GLP and non-GLP toxicology studies?

GLP studies follow a formal quality system (21 CFR Part 58 in the US, the OECD GLP Principles elsewhere) and are used as pivotal safety data. Non-GLP studies, such as dose range finding, are run earlier with less formality to choose doses and spot problems.

How long should repeat-dose toxicology studies be?

Under ICH M3(R2), repeated-dose toxicity studies should generally be at least as long as the clinical trial they support, for example a 2-week study for a trial of up to 2 weeks. Confirm the design for your program with a regulatory expert.

What is the safety pharmacology core battery?

ICH S7A defines it as studies of effects on the cardiovascular, respiratory and central nervous systems.

Why do biologics often use different species?

ICH S6(R1) asks for toxicology in species where the biologic is pharmacologically active, so species are chosen on target binding and cross-reactivity. One relevant species can be enough in justified cases.

What is a dose range finding study?

A short, usually non-GLP study at escalating doses that identifies tolerated doses and target organs, so the GLP study's doses can be chosen with confidence.

Is anything I type stored or shared?

No. The planner runs entirely in your browser. Nothing you enter is saved, sent to BioBridgeX, put in the page address or passed to analytics or AI services. Closing or refreshing the page clears it, so copy or print the plan before you leave.
